Synthesis Ordering

Get estimated quotes from DNA synthesis providers, then download your sequence and order on the provider's website.

Workflow

  1. Complexity screening — Nuclex checks your sequence for synthesis feasibility: GC content, homopolymers, repeats, hairpins, and local GC extremes.
  2. Biosafety gate — The design must pass biosafety screening before ordering. Flagged or blocked designs cannot be ordered.
  3. Quote comparison — Get estimated pricing from supported providers with best-price recommendation.
  4. Order from provider — Download your sequence and order directly on the provider's website. Log the order in Nuclex for your records.

Complexity checks

The complexity screen runs six checks:

  • GC content — overall GC must be between 25% and 65%
  • Homopolymers — runs of 6+ identical bases are flagged
  • Direct repeats — repeated segments that may cause recombination
  • Sequence length — must be within provider limits
  • Local GC extremes — 50 bp windows with <20% or >80% GC
  • Hairpin structures — inverted repeats that form secondary structures

Order logging

After placing an order on a provider's website, you can log it on the Orders page with your provider order ID. You can manually update the status as your order progresses through production, QC, and shipping.

Tier requirements

Synthesis quotes and FASTA download require the Researcher plan or higher. Free users can run complexity screens but cannot access quotes.
